A modern day adaptation of Cinderella with a twist as our Cinderella, Goo Hyo Sun, is loved by everyone whereas her stepsister, Eun Jo, has a dark past and struggles with the fact that her mother starts to care more for Hyo Sun than her.

Applying a modern twist to the classic fairy tale, the 2010 KBS drama Cinderella's Sister follows the contentious relationship between two stepsisters as their lives and loves intertwine. Tamra Island heroine Seo Woo is the Cinderella, while Korea's little sister Moon Geun Young grows up into the role of the older "evil stepsister". The two petite actresses deliver big performances as reluctant sisters whose polar personalities and love-hate relationship bring them through many ups and downs from adolescence to adulthood. Making his post-army comeback, Chun Jeong Myung adds to the angst and rivalry as the man they both love. Cinderella's Sister also co-stars veterans Lee Mi Sook and Kim Gab Soo, and 2PM's Ok Taec Yeon in his acting debut.

Since a young age, Eun Jo (Moon Geun Young : Painter of the wind, Innocent Steps) has learned to trust no one and live strong on her own. Used to being dragged from one broken home to another by her manipulative, money-grubbing mother (Lee Mi Sook), Eun Jo doesn't know what to make of her mother's latest catch Dae Sung (Kim Gab Soo), the solemn head of a traditional rice wine brewery. Both Dae Sung and his daughter, bubbly spoiled princess Hyo Sun (Seo Woo), warmly welcome wary young Eun Jo into their sprawling home, and treat her with a kindness that she simply can't stomach. Instead, she forms a bond with brewery staff Ki Hoon (Chun Jeong Myung), the first person to see past her tough act, and it is his betrayal that will hurt the most. Naive Hyo Sun, meanwhile, stubbornly seeks affection at every turn, only to slowly realize she's losing Ki Hoon and everything else in her life to her cold stepsister.
Song Eun-jo (Moon Geun Young), who lives with her mother since years ago (and also her mother's lover), finally gets a new family and a home when her mother remarries to a kind man who owns a winery. At first, she denied it, and even attempted to escape rather than dealing with the attempt of being schooled. The cynical Eun-jo is unable to accept the affection of her new step-sister Hyo-seon (Seo Woo) and step father Gu Dae-seong (Kim Gab Soo). Hyo-seon, with her bright character, is the apple in her father's eyes and Eun-jo's mother goes out of her way to fulfill the role of Hyo-seon's mother, causing her to neglect and act harshly towards Eun-jo.

However, Eun-jo finds a friend in Hong Ki-hoon (Chun Jung Myung), a college student working at the winery who becomes her own prince. Ki-hoon eventually leaves to take care of personal issues, but Hyo-seon's jealousy of Eun-jo causes her to make Eun-jo believe that he has abandoned her. The three reunite 8 years later to begin the main plot - a modern adaptation of Cinderella with a twist. Han Jeong-woo (Taecyeon), a boy that Eun-jo considered her little brother 8 years ago, appears in order to be near her and fulfills the role of a "fairy godmother" to her. A love triangle forms when Hyo-seon falls for Ki-hoon, who is still in love with Eun-jo. The feeling of betrayal lasts with Eun-jo, creating misunderstandings and difficulties. However, the kindness of Eun-jo's stepfather (Gu Dae-seong, Hyo-seon's father) begins to change everyone's lives, making this a tale of growth and redemption.
Gang-suk (played by Lee Mi-sook) and her daughter Eun-jo (played by Moon Geun-young) come to the Daeseongdoga brewery, a brewery of the traditional liquor "makgeolli." Gang-suk (known for her extensive experience with men) seduces the owner of the brewery, Gu Dae-seong (played by Kim Gab-soo), and the two get married.

Even though Hyo-seon (played by Seo Woo) loves her new stepmother and stepsister wholeheartedly, Eun-jo has trouble opening up her heart, even though she's a kindhearted, gentle soul deep down. Gi-hun (a helper who is like family, played by Chun Jung-myung) falls in love with Eun-jo and the three get caught in the ever-changing dynamics of love, hate, and misunderstanding. Eun-jo also has feelings for Gi-hun, but Gi-hun leaves for the military before he can learn Eun-jo's true feelings.

Eight years pass and Jeong-u (played by Taecyeon), the son of one of Gang-suk's many boyfriend's, comes back to find Eun-jo, who he fell in love with in his youth. Once an obese boy with few friends, he's now become a handsome and charming young man. Gi-hun also returns to Daeseongdoga around the same time, having completed his military service and studies abroad. During their absence, Eun-jo became even more involved in the family business, becoming a huge help to her stepfather. Eun-jo and Gi-hun (along with Hyo-seon) join forces to save the family business from Hongjuga, the nation's leading maker of traditional liquor who is trying to take over the brewery.

Gu Dae-seong, ignorant of the ties between Eun-jo and Gi-hun, believed that Gi-hun returned to Daeseongdoga to help the business; when he learns that Gi-hun is the illegitimate son of Hongjuga, he dies from a stroke. After his death, Gang-suk starts treating Hyo-seon with unusual coldness. As Hongjuga puts more and more pressure on Daeseongdoga, Eun-jo, Gi-hun, and Hyo-seon struggle to preserve the family brewery legacy. In the midst of it all, there hangs the unresolved issue of Jeong-u's love for Eun-jo, the love between Eun-jo and Gi-hun, and Hyo-seon's love for Gi-hun.
